Description
Complementary and alternative medicine are increasingly used by many to relief the effects of chronic stress. Odours derived from plants, like lavender are currently used to reduce stress. The main problem with traditional and new treatments is that they have not been properly tested with appropriate controls, existing evidence are predominately anecdotal. The main aim of this study was to properly evaluate the effectiveness of traditional and new plant derived odours in reducing the effects of chronic stress with appropriate controls. We used concurrent stressors of social isolation and predator odour as a model for chronically stressing mice over 10 days (D'Arbe et. al., 2002). Animals were divided into 4 groups (n=4 for each group): Group 1 controls (minimal/no stress) with odours, without odours group 2; stress with odours group 3 and stress without odours (group 4). We applied odours to only groups 1 and 3, three times a day by spraying 0.2 ml of odour onto bedding of the animal's cage. Following end of treatment, animals were sacrificed by cervical fracture, blood was collected from the heart and both vasa deferentia dissected free and place in an organ bath. We measured mean amplitudes of extra junctional currents (EJCs) using focal extracellular recording technique. At least 16 randomly chosen recording sites were electrophysiologically studied from each preparation. As demonstrated previously, stressed animals showed higher levels of sympathetic neurotransmission. We tested 7 different odours derived from green plants and graded their activity. We compared their activity with the commonly used stress relief agent, lavender and made an unexpected observation. The most effective plant derived odour, which reduced the physiological effects of chronic stress, was cis-3-hexen-1-ol [0.3%] followed by trans-2-hexenal [0.3%] and then by alpha-pinene [0.3%], the remaining odours had little or no effect. Lavender [0.3%] also had no effect. Our results and other published data suggest that lavender's stress relieving properties may be explained by its vasorelaxing effect (a cooling effect). We have identified 3 plant derived odours that significantly attenuate the stress induced upregulation of neurotransmission from sympathetic varicosities. Our study also demonstrates the highly plastic nature of neurotransmission efficacy.

1 The effects of stress in rats were evaluated by measuring changes in body weight and in responsiveness to noradrenaline (NA) in the isolated vas deferens and atria after the animals had been exposed to restraint or restraint and isolation. 2 Animals which were subjected to restraint alone (1 h day(-1) for 3, 7 or 28 days) had a significantly reduced rate of body weight gain. This effect was not potentiated by the additional stress of isolation. 3 Restraint alone did not produce significant changes in the responsiveness of the vas deferens to NA. However, adding isolation to restraint, as an additional stress, produced a further leftward shift of the NA dose-response curve for the vas deferens, so that there was a significant increase in sensitivity compared with control. 4 There was a significant rightward shift in the NA dose-response curves or reduction in sensitivity in the atria from animals which had been restrained for 7 or 28 days. Isolation did not produce a further rightward shift in the NA dose-response curve. 5 The results from this study indicate that the stress associated with repeated restraint reduces the rate of weight gain and reduces the responsiveness of the atria to NA. The responsiveness of the vas deferens to NA was increased by stress, but the combined effect of isolation and restraint was required to produce a significant effect. The differences in the effects of stress on these tissues could be associated with differences in presynaptic receptor populations.

Description
Current methods used to study neural communication have not been able to achieve both good spatial and temporal resolution of recordings. There are two ways to record synaptic potentials from nerve endings: recordings using single or dual intracellular or extra cellular metal electrodes give good temporal resolution but poor spatial resolution, and recording activity with fluorescent dyes gives good spatial resolution but poor temporal resolution. Such medical research activity in the area of neurological signal detection has thus identified a requirement for the design of a CMOS circuit that contains an array of independent sensors. As both spatial and temporal distribution of acquired data is required in this application, the circuit must be capable of continuous measurement of synaptic potentials from an array of points on a tissue sample, with a 10 µm separation between sensor points.The major requirement for the circuit is that it is capable of sensing synaptic potentials of the order of several mV, with a resolution of 0.05 mV. For data recording purposes, the circuit must amplify these synaptic potentials and digitise them together with their locations in the sensor array. Finally, the circuit must be biologically inert, to avoid specimen deterioration.This paper presents the design of a prototype single-chip circuit, which provides a 6 x 3 array of independent synaptic potential sensors. The signal from each of the sensors is amplified and time-multiplexed into an on-chip A/D converter. The circuit provides an 8-bit synaptic potential value, together with an 8-bit field containing array location and trigger signals suitable for external data acquisition instrumentation.Our test circuit is implemented in a low-cost 0.5 um, 5 V CMOS process. The fabricated die is mounted in a standard 40 pin DIP ceramic package, with no lid to allow direct contact of the die surface with the tissue sample. The only post-processing step required for these packages is to encapsulate the exposed bond wires to ensure that the device is biologically inert. No further processing of the silicon die is required. Both the circuit design and the chip performance will be presented in the seminar.
